    Testing Eurasian wild boar piglets for serum antibodies against Mycobacterium bovis

Che’ Amat et al.Animal tuberculosis (TB) caused by infection with Mycobacterium bovis and closely related members of the M. tuberculosis complex (MTC), is often reported in the Eurasian wild boar (. Sus scrofa). Tests detecting antibodies against MTC antigens are valuable tools for TB monitoring and control in suids. However, only limited knowledge exists on serology test performance in 2-6 month-old piglets. In this age-class, recent infections might cause lower antibody levels and lower test sensitivity. We examined 126 wild boar piglets from a TB-endemic site using 6 antibody detection tests in order to assess test performance. Bacterial culture (. n=. 53) yielded a M. bovis infection prevalence of 33.9%, while serum antibody prevalence estimated by different tests ranged from 19% to 38%, reaching sensitivities between 15.4% and 46.2% for plate ELISAs and between 61.5% and 69.2% for rapid immunochromatographic tests based on dual path platform (DPP) technology. The Cohen kappa coefficient of agreement between DPP WTB (Wildlife TB) assay and culture results was moderate (0.45) and all other serological tests used had poor to fair agreements. This survey revealed the ability of several tests for detecting serum antibodies against the MTC antigens in 2-6 month-old naturally infected wild boar piglets. The best performance was demonstrated for DPP tests. The results confirmed our initial hypothesis of a lower sensitivity of serology for detecting M. bovis-infected piglets, as compared to older wild boar. Certain tests, notably the rapid animal-side tests, can contribute to TB control strategies by enabling the setup of test and cull schemes or improving pre-movement testing.     Incremental learning framework-based condition monitoring for novelty fault identification applied to electromechanical systems

    A great deal of investigations are being carried out towards the effective implementation of the 4.0 Industry new paradigm. Indeed, most of the machinery involved in industrial processes are intended to be digitalized aiming to obtain enhanced information to be used for an optimized operation of the whole manufacturing process. In this regard, condition monitoring strategies are being also reconsidered to include improved performances and functionalities. Thus, the contribution of this research work lies in the proposal of an incremental learning framework approach applied to the condition monitoring of electromechanical systems. The proposed strategy is divided in three main steps, first, different available physical magnitudes are characterized through the calculation of a set of statistical-time based features. Second, a modelling of the considered conditions is performed by means of self-organizing maps in order to preserve the topology of the data; and finally, a novelty detection is carried out by a comparison among the quantization error value achieved in the data modelling for each of the considered conditions. The effectiveness of the proposed novelty fault identification condition monitoring methodology is proved by means of the evaluation of a complete experimental database acquired during the continuous working conditions of an electromechanical system. © 2018 IEEE.Peer ReviewedPostprint (author's final draft

    Novelty detection based condition monitoring scheme applied to electromechanical systems

    This study is focused on the current challenges dealing with electromechanical system monitoring applied in industrial frameworks, that is, the presence of unknown events and the limitation to the nominal healthy condition as starting knowledge. Thus, an industrial machinery condition monitoring methodology based on novelty detection and classification is proposed in this study. The methodology is divided in three main stages. First, a dedicated feature calculation and reduction over each available physical magnitude. Second, an ensemble structure of novelty detection models based on one-class support vector machines to identify not previously considered events. Third, a diagnosis model supported by a feature fusion scheme in order to reach high fault classification capabilities. The effectiveness of the fault detection and identification methodology has been compared with classical single model approach, and verified by experimental results obtained from an electromechanical machine. © 2018 IEEE.Postprint (author's final draft

    Fec, CA5A and CLSTN2 genes and their function during sheep ovulation: a review.

    Objective: To describe the function of Fec, CA5A and CLSTN2 genes during ovulation in ewes. Design/methodology/approach: A search and analysis of scientific information related to Fec, CA5A and CLSTN2 genes in sheep was performed. Results: Fec, CA5A and CLSTN2 genes are involved at the ovarian level; ewes carrying the first gene were found to have increased ovulation rate, folliculogenesis and granulosa cell differentiation. CA5A stimulates an increased follicular rate and plays an important role in pre-implantation. While CLSTN2 has activity in ovarian development and growth; it also has the ability to interact with other genes involved in follicular maturation, granulosa cell differentiation and development of the ovarian follicle. Limitations on study/implications: Ewes carrying these genes increase the prolificacy rate in the flock. Findings/conclusions: The expression of these genes acts synergistically in the ovulatory process, enhancing the ovulatory response by contributing to endocrine, paracrine and molecular synchronisation, so that the maturation of the oocyte occurs, leading to ovulation

    An industrial machinery condition monitoring methodology based on ensemble novelty detection and evolving classification is proposed in this study. The methodology contributes to solve current challenges dealing with classical electromechanical system monitoring approaches applied in industrial frameworks, that is, the presence of unknown events, the limitation to the nominal healthy condition as starting knowledge, and the incorporation of new patterns to the available knowledge. The proposed methodology is divided into four main stages: 1) a dedicated feature calculation and reduction over available physical magnitudes to increase novelty detection and fault classification capabilities; 2) a novelty detection based on the ensemble of one-class support vector machines to identify not previously considered events; 3) a diagnosis by means of eClass evolving classifiers for patterns recognition; and 4) re-training to include new patterns to the novelty detection and fault identification models.     Assessment of an oral Mycobacterium bovis BCG vaccine and an inactivated M. bovis preparation for wild boar in terms of adverse reactions, vaccine strain survival, and uptake by nontarget species

    Wildlife vaccination is increasingly being considered as an option for tuberculosis control. We combined data from laboratory trials and an ongoing field trial to assess the risk of an oral Mycobacterium bovis BCG vaccine and a prototype heat-inactivated Mycobacterium bovis preparation for Eurasian wild boar (Sus scrofa). We studied adverse reactions, BCG survival, BCG excretion, and bait uptake by nontarget species. No adverse reactions were observed after administration of BCG (n = 27) or inactivated M. bovis (n = 21). BCG was not found at necropsy (175 to 300 days postvaccination [n = 27]). No BCG excretion was detected in fecal samples (n = 162) or in urine or nasal, oral, or fecal swab samples at 258 days postvaccination (n = 29). In the field, we found no evidence of loss of BCG viability in baits collected after 36 h (temperature range, 11°C to 41°C). Camera trapping showed that wild boar (39%) and birds (56%) were the most frequent visitors to bait stations (selective feeders). Wild boar activity patterns were nocturnal, while diurnal activities were recorded for all bird species. We found large proportions of chewed capsules (29%) (likely ingestion of the vaccine) and lost baits (39%) (presumably consumed), and the proportion of chewed capsules showed a positive correlation with the presence of wild boar. Both results suggest proper bait consumption (68%). These results indicate that BCG vaccination in wild boar is safe and that, while bait consumption by other species is possible, this can be minimized by using selective cages and strict timing of bait deployment.This is a contribution to Plan Nacional I+D+i AGL2011-30041 from the Ministerio de Economía y Competitividad (MINECO), Spain, and FEDER and to EU FP7 grant WildTBvac.     Comparative genomics of field isolates of Mycobacterium bovis and M. caprae provides evidence for possible correlates with bacterial viability and virulence

The life cycle of mycobacteria is complex and the mechanisms resulting in pathogen infection and survival in host cells are not fully understood. Recently, comparative genomics analyses have provided new insights into the evolution and adaptation of the MTBC to survive inside the host. However, most of this information has been obtained using M. tuberculosis but not other members of the MTBC such as M. bovis and M. caprae. In this study, the genome of three M. bovis (MB1, MB3, MB4) and one M. caprae (MB2) field isolates with different lesion score, prevalence and host distribution phenotypes were sequenced. Genome sequence information was used for whole-genome and protein-targeted comparative genomics analysis with the aim of finding correlates with phenotypic variation with potential implications for tuberculosis (TB) disease risk assessment and control. At the whole-genome level the results of the first comparative genomics study of field isolates of M. bovis including M. caprae showed that as previously reported for M. tuberculosis, sequential chromosomal nucleotide substitutions were the main driver of the M. bovis genome evolution. The phylogenetic analysis provided a strong support for the M. bovis/M. caprae clade, but supported M. caprae as a separate species. The comparison of the MB1 and MB4 isolates revealed differences in genome sequence, including gene families that are important for bacterial infection and transmission, thus highlighting differences with functional implications between isolates otherwise classified with the same spoligotype. Strategic protein-targeted analysis using the ESX or type VII secretion system, proteins linking stress response with lipid metabolism, host T cell epitopes of mycobacteria, antigens and peptidoglycan assembly protein identified new genetic markers and candidate vaccine antigens that warrant further study to develop tools to evaluate risks for TB disease caused by M. bovis/M.caprae and for TB control in humans and animals.This research was supported by grants AGL2014-56305 and IPT-2011-0735-010000 from Ministerio de Economía y Competitividad, Spain, and the European Union FP7 ANTIGONE grant 278976 and Horizon 2020 COMPARE Grant 377/14.Peer Reviewe

    A plasma fatty acid profile associated to type 2 diabetes development: from the CORDIOPREV study

    Purpose: The prevalence of type 2 diabetes mellitus (T2DM) is increasing worldwide. For this reason, it is essential to identify biomarkers for the early detection of T2DM risk and/or for a better prognosis of T2DM. We aimed to identify a plasma fatty acid (FA) profile associated with T2DM development. Methods: We included 462 coronary heart disease patients from the CORDIOPREV study without T2DM at baseline. Of these, 107 patients developed T2DM according to the American Diabetes Association (ADA) diagnosis criteria after a median follow-up of 60 months. We performed a random classification of patients in a training set, used to build a FA Score, and a Validation set, in which we tested the FA Score. Results: FA selection with the highest prediction power was performed by random survival forest in the Training set, which yielded 4 out of the 24 FA: myristic, petroselinic, α-linolenic and arachidonic acids. We built a FA Score with the selected FA and observed that patients with a higher score presented a greater risk of T2DM development, with an HR of 3.15 (95% CI 2.04–3.37) in the Training set, and an HR of 2.14 (95% CI 1.50–2.84) in the Validation set, per standard deviation (SD) increase. Moreover, patients with a higher FA Score presented lower insulin sensitivity and higher hepatic insulin resistance (p < 0.05). Conclusión: Our results suggest that a detrimental FA plasma profile precedes the development of T2DM in patients with coronary heart disease, and that this FA profile can, therefore, be used as a predictive biomarker

    Laboratory diagnosis of severe hypertriglyceridaemia. Cases from the dyslipidaemia regristy of the spanish atherosclerosis society

    Background and Aims Severe hypertriglyceridaemia (sHTG) increases the risk of cardiovascular disease and acute pancreatitis episodes. Patients with sHTG fit mainly into two clinical entities: Familial or Multifactorial Chylomicronemia Syndromes (FCS and MCS, respectively). FCS and MCS exhibit clinical differences but also separate genetic and biochemical characteristics that can be assessed in the laboratory. The aim of this work has been to implement a laboratory workflow to help diagnose sHTG patients with either FCS or MCS. Methods Patients with two fasting triglycerides >1000mg/dL determinations were sequenced with a capture probe panel of 24 triglycerides-related genes using massive parallel sequencing (n=200). Two-step sequential ultracentrifugation was performed (n= 159) to diagnose Type I hyperlipoproteinemia (HLP I) and post heparin lipoprotein lipase activity was measured to discard or confirm its deficiency (n=60). Results Most patients had MCS as they: (i) did not exhibit HLPI and/or (ii) their genetic profile was not compatible with FCS and (iii) were not deficient in LPL activity. FCS cases were identified as they had: (i) HLPI, and/or (ii) biallelic pathogenic variants in LPL (n=5), GPIHBP1 (n=3), or LMF1 (n=2) genes and/or (iii) LPL activity deficiency. We identified 4 FCS patients with HLPI, biallelic pathogenic variants in APOA5 but a rescued LPL activity. An additional study of Apo-AV functionality was designed to confirm the FCS diagnosis in these cases. Conclusions Laboratory studies, in patients with severe hypertriglyceridaemia, provide with information of clinical utility to distinguish between Familial and Multifactorial Chylomicronemia Syndromes.Universidad de Málaga.
